Comment:

Target is the 6 kb deletion junction in the maize waxy gene locus.

Based on NCBI Reference Sequence: NC_050104.1 (Zm-B73-REFERENCE-NAM-5.0, whole genome shotgun sequence) forward primer P3 includes a 3 bp insertion (indicated in P3 as small letters) not complementary to the reference sequence. This might be explained by differences in the PH184C inbred line (mother line for GE-CRISPR-Cas Waxy Corn) compared to the B73 reference genome.
