Spodoptera frugiperda OX5382G carries a conditional female lethality trait (1) and a fluorescent marker (2).
- Repressible female specific lethality is achieved using an alternative splicing module of the Spodoptera frugiperda doublesex gene (dsx) and the expression of a synthetic tetracycline‐transcriptional activator (tTAV) variant. In the environment, without the presence of tetracycline antibiotics, female offspring of the OX5382 fall armyworm are expected to die before they mature into adults. Due to the alternative splicing mechanism, males carry and transmit the self-limiting genes to progeny without requiring tetracycline. Population suppression is thus achieved through the biased selection of male offspring
- The expressed fluorescent marker DsRed2, a synthetically modified variant of the original red fluorescent protein isolated from Discosoma sp., aids in the detection of Spodoptera frugiperda carrying the OX5382 rDNA construct. A synthetic nuclear localisation signal was included to the expression cassette to localise DsRed2 to the nuclei of cells.
