GE-PPO2 Potato was developed by PEG-mediated protoplast transfection of the Cas9-ribonucleoprotein (RNP) delivery system. Mediated by a single gRNA the ppo2 gene was edited. ppo2 encodes for the polyphenol oxidase PPO, which is associated with the enzymatic browning process in potato tubers.
Two lines #38 and #165 were selected based on the PPO enzyme activity and decreased browning. In both lines, deep-Seq analysis of the target sites in the ppo2 gene shows specific deletions in all four alleles of 2 bp (line #38) or 4 bp (line #165).
During development of these two lines no foreign DNA sequences were introduced in the plant genome. Modifications in the iso-genes ppo1, ppo3, and ppo4 were not observed.
In the context of the RSR request the developer does not specify if line #38 or line #165 is favored as the final event.
