The GE-eBSV Banana events were generated by editing the integrated endogenous banana streak virus (eBSV) sequences in "Gonja Manjaya", a false horn plantain.
The eBSV activates into infectious viral particles under stress and the plant develops disease symptoms.
CRISPR/Cas9-based genome-editing technology was applied to inactivate the eBSV in the B genome
of plantain (AAB genome). The CRISPR/ Cas9 construct, with three gRNAs targeting the three ORFs of eBSV, was delivered into the embryogenic cell suspensions of Gonja Manjaya via Agrobacterium tumefaciens.
The GE-eBSV banana events showed mutations, indels of 1 bp up to 198 bp in the targeted sites, with the potential to prevent proper transcription or/and translation into functional viral proteins.
The stability of the mutations was shown for one event over multiple rounds of vegetative propagation.
